I think you are looking for something some third party tools do provide. You might search for "BoxCryptor", which is able to create a layer "above" OneDrive and transparently encrypt/decrypt the files before saving/loading the files. This way the files on the disk and in the cloud are encrypted and will be decrypted "on the fly" when accessing them. By integrating such a system into OneDrive, the files in the cloud might also be accessible without the software, but by providing the key to the browser.
If you want just to protect the files on your PC you might use some kind of full disk encryption like BitLocker (win 10 pro) or some 3rd party software. -
